AOD-9604 is a modified fragment of growth hormone (hGH) encompassing amino acids 176-191 from the C-terminal region, with an added tyrosine residue at the N-terminus. This fragment was isolated based on research identifying the lipolytic domain of growth hormone as distinct from its growth-promoting and diabetogenic regions. The peptide retains the fat-metabolizing activity attributed to the full hGH molecule while lacking the growth-promoting effects mediated by IGF-1, making it a subject of research into targeted fat metabolism modulation.
Preclinical studies in obese animal models have demonstrated that AOD-9604 stimulates lipolysis (fat breakdown) and inhibits lipogenesis (fat formation) without affecting blood glucose or tissue growth. The mechanism is believed to involve activation of beta-3 adrenergic receptor-mediated pathways in adipose tissue, mimicking the lipolytic cascade triggered by growth hormone without the associated insulin resistance. Research by Heffernan et al. demonstrated that chronic administration in obese Zucker rats produced significant reductions in body fat mass without changes in lean body mass or food consumption.
